چکیده

Platelets are highly specialized blood cells critically involved in hemostasis and thrombosis. Members of the protein kinase C (PKC) family have established roles in regulating platelet function and thrombosis, but the molecular mechanisms are not clearly understood. In particular, the conventional PKC isoform, PKC , is a major regulator of platelet granule secretion, but the molecular pathway from PKC to secretion is not defined. Protein kinase D (PKD) is a family of 3 kinases activated by PKC, which may represent a step in the PKC signaling pathway to secretion. In the present study, we show that PKD2 is the sole PKD member regulated downstream of PKC in platelets, and that the conventional, but not novel, PKC isoforms provide the upstream signal. Platelets from a gene knock-in mouse in which 2 key phosphorylation sites in PKD2 have been mutated (Ser707Ala/Ser711Ala) show a significant reduction in agonist-induced dense granule secretion, but not in -granule secretion. This deficiency in dense granule release was responsible for a reduced platelet aggregation and a marked reduction in thrombus formation. Our results show that in the molecular pathway to secretion, PKD2 is a key component of the PKC-mediated pathway to platelet activation and thrombus formation through its selective regulation of dense granule secretion. Regular Article PLATELETS AND THROMBOPOIESIS PDK1 regulates platelet activation and arterial thrombosis

Platelets, which are derived from megakaryocytes, circulate in mammalian blood and play essential roles in hemostasis, angiogenesis, inflammation, and metastasis. Phosphoinositide 3-kinases (PI3Ks) are a conserved family of enzymes, each having both protein and lipid kinase activities. PI3K-mediated signaling affects platelet adhesion and aggregation. PI3Ks are activated downstream of several m...

PLATELETS AND THROMBOPOIESIS AMPK 2 subunit is involved in platelet signaling, clot retraction, and thrombus stability

The adenosine monophosphate (AMP)– activated protein kinase (AMPK) is a regulator of energy balance at the cellular and whole-body levels, but little is known about the role of AMPK in platelet activation. We report that both the 1 and 2 AMPK isoforms are expressed by human and murine platelets and that thrombin elicits the phosphorylation of AMPK as well as the upstream kinase, liver kinase B1...

PLATELETS AND THROMBOPOIESIS CalDAG-GEFI is at the nexus of calcium-dependent platelet activation

The importance of the second messengers calcium (Ca2 ) and diacylglycerol (DAG) in platelet signal transduction was established more than 30 years ago. Whereas protein kinase C (PKC) family members were discovered as the targets of DAG, little is known about the molecular identity of the main Ca2 sensor(s). We here identify Ca2 and DAG-regulated guanine nucleotide exchange factor I (CalDAG-GEFI...

PLATELETS AND THROMBOPOIESIS Transforming growth factor- –induced protein (TGFBIp/ ig-h3) activates platelets and promotes thrombogenesis

Transforming growth factor–induced protein (TGFBIp)/ ig-h3 is a 68-kDa extracellular matrix protein that is functionally associated with the adhesion, migration, proliferation, and differentiation of various cells. The presence of TGFBIp in platelets led us to study the role of this protein in the regulation of platelet functions. Upon activation, platelet TGFBIp was released and associated wit...

PLATELETS AND THROMBOPOIESIS S6K1 and mTOR regulate Rac1-driven platelet activation and aggregation

Platelet activation and thrombus formation are under the control of signaling systems that integrate cellular homeostasis with cytoskeletal dynamics. Here, we identify a role for the ribosome protein S6 kinase (S6K1) and its upstream regulator mTOR in the control of platelet activation and aggregate formation under shear flow. Platelet engagement of fibrinogen initiated a signaling cascade that...
